This may be as
expected, as the most basic building block of organic chemistry is
the transfer of single carbon groups. From building the bases of DNA
to putting the final touches on complex protein structures, single
carbon transfers are the brick and mortar of organic chemistry.
The simplest
organic molecule contains a single carbon surrounded by 4 atoms of
hydrogen. Called methane, it has the shape of a perfect 4-sided
pyramid called a tetrahedron. In this structure, the carbon is in

If a hydrogen atom
is removed from one corner of the methane pyramid, the carbon atom

which life as we know it would be impossible.
One of the most
important methyl group transfers in the body is onto DNA itself. Of
the 4 DNA bases, only one base — cytosine — is the recipient of
methyl groups (on the number 5 position of its ring structure to
make it 5-methyl-cytosine). The process of adding a methyl group to
a molecule is called methylation; removing a methyl group is called
demethylation; restoring a methyl group that has been removed to its
biologically desirable location is termed remethylation.
When a human ovum
is fertilized, an enormously complex task of development and
differentiation ensues to give rise to an infant with a multitude of
different cells and tissues. The body uses methyl groups as anchors
on the DNA itself to organize the function of every type of cell.
Attaching to the DNA base cytosine, the specific pattern of DNA
methylation is unique for every cell. The DNA methylation pattern
tells each cell what part of the DNA can be active and which should
be dormant.
When an infant is
born, the methylation of cytosine ranges from 2-6%, the fraction
dependent on the unique pattern for each cell type. This pattern is
like a fingerprint for each cell and gives the cell a blueprint for
which proteins and enzymes and activities it will carry into
function.
Using an estimated
average of 3% methylation of cytosine at birth, the nuclear DNA of
each cell would have approximately 90 million methyl groups. As
reviewed by a recent article in the journal Medical Hypothesis (Vol.
51, pages 179-221, 1998), the body controls development and aging by
the loss of methyl groups from DNA. Death from degenerative causes
usually occurs when about 40% of the methyl groups have been lost
from DNA.
Cellular
information and control systems become increasingly fuzzy as DNA
methyl groups are lost. Certain cancers tend to occur at a 20% DNA
methyl group loss. In autoimmune diseases like lupus and rheumatoid
arthritis, a specific immune cell called the lymphocyte has been
shown to lose 50% of its DNA methyl groups. Loss of information
integrity in this powerful type of immune cell causes an attack of
self against self, as lymphocytes attack otherwise normal, healthy
tissues.
An average loss of
roughly 36 million DNA methyl groups per cell represents a limit to
biological survival. An average loss of 1800 DNA methyl groups per
cell per day limits a lifespan to about 60-65 years, whereas a
reduced loss to only 1200 groups per cell per day is consistent with
survival to age 90-95.
Factors that
accelerate methyl group loss are smoking, excessive alcohol intake,
vitamin deficiencies, synthetic female hormones, and lack of
adequate exercise.
